چکیده

Background Stromal derived factor (SDF-1) is a natural ligand for CXCR4 and chemokine (C-C) motif ligand 3-like 1 (CCL3L1) is for CCR5 HIV coreceptors. The individual role of the SNP in 3′ untranslated region of SDF-1 (SDF1-3′A) and low copy number of the CCL3L1 gene in determining susceptibility to HIV infection is well documented. The aim of the present study was to analyze the synergistic effect of the SNP in SDF-1 gene and CNV in CCL3L1 gene influencing the susceptibility to HIV-1/AIDS in Indians.

The influence of CCL3L1 gene-containing segmental duplications on HIV-1/AIDS susceptibility.

Segmental duplications in the human genome are selectively enriched for genes involved in immunity, although the phenotypic consequences for host defense are unknown. We show that there are significant interindividual and interpopulation differences in the copy number of a segmental duplication encompassing the gene encoding CCL3L1 (MIP-1alphaP), a potent human immunodeficiency virus-1 (HIV-1)-...

CCL3L1 Copy Number Variation and Susceptibility to HIV-1 Infection: A Meta-Analysis

BACKGROUND Although several studies have investigated whether CCL3L1 copy number variation (CNV) influences the risk of HIV-1 infection, there are still no clear conclusions. Therefore, we performed a meta-analysis using two models to generate a more robust estimate of the association between CCL3L1 CNV and susceptibility to HIV-1 infection. METHODS We divided the cases and controls into two ...

Influence of variations in CCL3L1 and CCR5 on tuberculosis in a northwestern Colombian population.

We investigated the association of polymorphisms in CCR5, the major human immunodeficiency virus (HIV)-1 coreceptor, and copy number of its potent ligand CCL3L1 with tuberculosis in 298 individuals from Colombia. The CCR5-HHD haplotype, a known genetic determinant of increased susceptibility to HIV-AIDS, and a high copy number of CCL3L1, a known genetic determinant of enhanced CCL3/CCL3L1 chemo...

CCL3L1-CCR5 Genotype Improves the Assessment of AIDS Risk in HIV-1-Infected Individuals

BACKGROUND Whether vexing clinical decision-making dilemmas can be partly addressed by recent advances in genomics is unclear. For example, when to initiate highly active antiretroviral therapy (HAART) during HIV-1 infection remains a clinical dilemma. This decision relies heavily on assessing AIDS risk based on the CD4+ T cell count and plasma viral load. However, the trajectories of these two...

CCL3L1 copy number, HIV load, and immune reconstitution in sub-Saharan Africans

BACKGROUND The role of copy number variation of the CCL3L1 gene, encoding MIP1α, in contributing to the host variation in susceptibility and response to HIV infection is controversial. Here we analyse a sub-Saharan African cohort from Tanzania and Ethiopia, two countries with a high prevalence of HIV-1 and a high co-morbidity of HIV with tuberculosis. METHODS We use a form of quantitative PCR...
